The preservation of sperm genomic integrity is critical for successful fertilization and subsequent embryonic development following artificial insemination. Cryopreservation-associated oxidative stress can compromise sperm DNA and may ultimately reduce reproductive efficiency. The present study evaluated the effect of platelet-rich plasma (PRP) supplementation on sperm DNA integrity and assessed its potential reproductive benefit under field conditions. Goat semen ejaculates were extended with TRIS egg yolk citrate extender supplemented with 0% PRP (control), 5% PRP (T1), 10% PRP (T2), and 15% PRP (T3), followed by cryopreservation and storage in liquid nitrogen (−196°C) for 72 h. Post-thaw samples were evaluated for sperm DNA fragmentation, and the treatment demonstrating the most favorable overall sperm response was subsequently evaluated through field artificial insemination trials. Spermatozoa supplemented with 10% PRP exhibited significantly lower DNA fragmentation compared with the control and other treatment groups, indicating enhanced preservation of genomic integrity during cryopreservation. In the field trial, artificial insemination using PRP-supplemented semen resulted in a conception rate of 41.52%, compared with 38.03% in the control group. The concurrent improvement in sperm DNA integrity and conception outcome suggests that PRP-mediated cellular protection may contribute to improved reproductive competence of cryopreserved spermatozoa. These findings provide a link between preservation of sperm genomic integrity and field reproductive performance and highlight 10% PRP as a promising biological additive for enhancing the quality and fertility potential of cryopreserved goat semen.
